2017) allow the observation of a cryo-fixed specimen in a more advanced LM at better resolution and in an EM with high acceleration voltage. 2016) and Leica cryo-CLEM System (Hampton et al. In comparison, separated CLEM systems such as the CorrSight system equipped with a cryo module from Thermo Fisher Scientific Inc. The iCorr system thus can only have fluorescence signals at the green channel with a low resolution of about 500 nm (Wang et al. Within the iCorr system, however, only small light objective lens as 15×/0.5 NA can be integrated due to the limited space between the pole pieces of a standard Tecnai Spirit TEM. The experiments efficiency is greatly increased without repeated grid transfers (Agronskaia et al. The first integrated CLEM system called Tecnai with iCorr (Thermo Fisher Scientific Inc.) allows the stage to be tilted 90° to switch between the LM and EM mode for direct correlation of a specimen in situ, preventing the specimen from potential contamination and damage. 2010).įor the second type of CLEM method, two different designs are commercially available. While the first type of CLEM method provides convenience of use and better resolution of LM, the second type guarantees more accurate correlation and is more favored (Briegel et al. 2010 Schorb and Briggs 2014 Schorb et al. Special specimen stages designed for the related instruments are essential to preserve the cryo-fixed specimens (Li et al. Such a practice needs the cryo-fixed specimen to be maintained always in a humidity-free environment and below ~−140 ☌ during the whole CLEM process to prevent ice contamination on the specimen surface or recrystallization in the specimen. The sample is plunge frozen or cryo-sectioned first, then the grid is visualized under LM and EM sequentially with little disturbance of its structure, thus making the correlation more consistent. (2) Correlative LM/EM with freezing before FM imaging (Briegel et al. Therefore, it works better for specimen that is relatively stable and the fluorescently marked structure would not change in the seconds to minutes before vitrification. Various factors can be introduced to the specimen between the LM imaging and specimen vitrification, causing more difficulties for accurate correlation. After FM imaging, the sample needs to be frozen for further cryo-EM imaging. FM can have better resolution with high-powered, oil-immersion lenses with large numerical apertures. FM imaging of samples at room temperature could be done with any typical FM instrument. Currently, two types of methods are used for correlation: (1) correlative LM/EM with freezing after FM imaging (Briegel et al. Cryo-CLEM is thus developed in order to visualize vitrified specimens by cryo-EM (Bykov et al. 2014), vitrified biological specimens become more and more popular to avoid the potential artifacts and damages caused by the chemical fixation. 2016 Kong and Loncarek 2015 Loussert Fonta and Humbel 2015 Mourik et al. While the chemical fixed specimens are still the majority of targets investigated by CLEM (Hellstrom et al. 2012).ĬLEM technique was first reported in the early 1960s for adenovirus study with conventional chemical fixation EM specimen preparation (Godman et al. The advantages of specific labeling and localization in LM and high spatial resolution in EM can be combined, leading to the development of a powerful technique as the correlative light and electron microscopy (CLEM) (Anderson et al. Light microscopy (LM), more specifically, fluorescence microscopy (FM) is well developed to label specific molecules and localize them in a large viewing area with a resolution of several hundreds of nanometers. Furthermore, the large dimension of a cell and a rather small viewing area of EM at a high magnification render it more difficult to localize relatively sparse molecules in a specific cellular structure environment (Plitzko et al. The major hurdle lies in the fact that many different kinds of macromolecules are crowded within the cell and therefore are hard to be distinguished from each other simply by their shapes revealed in cryo-EM (Bauerlein et al. 2008 Oikonomou and Jensen 2017 Plitzko et al. However, it is still very challenging to identify and determine the structures of interested macromolecules in situ with cryo-electron microscopy (cryo-EM) (Lucic et al. In combination with the most recent hardware and software breakthroughs, cryo-fixed macromolecule complexes isolated from cells can be determined at near atomic resolution using the single particle reconstruction method (Ma et al. Vitrification of biological specimens in liquid nitrogen (LN2) temperature has been proved to be a powerful technique to preserve the native structures of macromolecules either in vitro or in situ.

3/30/2024 0 Comments New york scratch off codesNo dice – they just told me to contact the Lottery Commission if I wanted to complain.Īnd I did. But the cashier didn’t care how many times I pointed out that hey, this is technically a win – look right there! These markers show the I won the game per the rules of blackjack. I had this happen to me once with a scratcher game modeled after the card game of blackjack. The first thing they will do is scan the code – no ding? No bling. When you scratch off all the little squares on a scratch game, you may find that hey, this is a win! Look at the line in bingo, or the three cherries, or the matching bells – this is a win! Take it to the store and give it to the cashier. If It’s Not a Winning Code, You’re Not a Winner The best strategy then is to buy tickets in bunches, in a number statistically offering the best chance of buying a winner without spending more than necessary. Assuming you spotted such a flaw, you would have to convince a clerk to let you examine the tickets – normally hidden under the counter – before buying them. However, he spotted a flaw in the game design on the face of the scratch game, not the letter code itself. The story of a statistician who spotted a pattern on the scratchers in Ontario, Canada, published in Wiredin 2011, is often repeated as “cracking the code” to the lottery. No clerk would let you scratch off the code before buying. However, can you see the code before you buy it? No, usually the code is hidden under the scratch-off layer, along with the bar code itself. Of course! That’s the point of the code, to indicate with no doubt that this is one of the remaining prizes. Ensuring Fair Play: The use of validation codes promotes fairness by guaranteeing that only tickets genuinely issued by the lottery can be redeemed for prizes. This protects both players and the integrity of the lottery. Preventing Counterfeits: By concealing the code beneath the scratch-off material, counterfeiters are deterred from creating fake tickets. They act as a safeguard against fraudulent activities, ensuring that only legitimate tickets are redeemed. Security and Authenticity: Validation codes play a critical role in maintaining the security and authenticity of scratcher tickets. The lottery retailer or official can then scan or manually input this code into the lottery system for validation. Verification Process: When a player wins, they can redeem their prize by presenting the scratcher with the intact validation code.If someone attempts to alter the code, the scratcher becomes void, alerting both the player and the lottery. Tamper-Proofing: Validation codes are hidden under the scratch-off material to deter counterfeiters. This uniqueness is crucial in preventing fraud and ensuring fair play. Unique Identifiers: Each scratcher ticket possesses a distinct validation code that distinguishes it from others. But these codes aren’t always so comprehensible – you should always scan the bar code to confirm it’s a winner than attempt to interpret these codes. For example, “OHM” indicates $100,000 (O = one, H = hundred, M = “milieu” or thousand in French). This article on Colorado lottery scratcher codes shows a list of codes that indicate the amount of the prize. Presumably, this is also for a machine to clearly read the numbers in a machine-readable format, not obscured by font styles and art design. Some websites compThey are rough translations of the winning amount, if that ticket is a winner. Many people online point out the “secret” of these codes. The validation code: the letters printed around the same area as the bar code. This machine connects to the Lottery Commission’s database of winning tickets. The bar code: the cashier will scan these into their lottery scanner machine, which will either ring bells if it’s a winner or do nothing if a loser. Some may indicate the prize amount, ticket status, or other relevant information. These codes are encrypted in a way that allows lottery systems to quickly verify their authenticity. These codes are usually hidden beneath the latex scratch-off surface, ensuring they are revealed only when the ticket is scratched. It serves as a vital identifier, helping lottery operators and players verify the authenticity and validity of the ticket. What Are Validation Codes?Ī validation code is a unique alphanumeric sequence assigned to each scratcher ticket by the lottery. In this article, we delve into what validation codes are, how they work, and why they matter to both players and the lottery operators. Besides the immediate gratification of revealing a prize, each scratcher ticket contains something less visible but equally important: a validation code. The excitement of scratching off a ticket to reveal if you’re a winner is a thrill many enjoy. 3/30/2024 0 Comments Tunnel rush online unblockedPlaying Tunnel Rush unblocked chrome game can be a fun and enjoyable way to relieve stress and improve mental well-being. In a video where he claims to be rebutting lies, he manages to spread even more lies, like stating SCVI was a huge success because even the very popular SCII had also sold only 2M. And we can't even say he's particularly good at assessing the information that is out there. If you see his videos from one year ago, he also made it seem like S3 was a given, then it was a maybe, finally he 'confirmed' (his words) it wasn't happening. Soulcalibur IV is the sixth entry in Namco Bandai’s Soul series. He doesn't have any inside info, so all he says is personal opinion and speculation, even if he seems to convey those as a fact.Īlthough I think it's positive for SC to have a YT channel as dedicated and passionate as his, the reason I won't even bother with his videos anymore is exactly that, whether he's aware of it or not, he kinda passes his ramblings as facts. Where SoulCalibur IV and V tried to add too many mechanics and systems at once, SoulCalibur VI holds back. If SCVI didn't turn a profit, then I'd really need to see the financials for the game as well as for several other fighting series that keep popping games despite their sales coming not even close to 1M, because otherwise I definitely will never understand.Ībout this Mark Yoon fellow. SCVI was generally well received and it was obviously a budget title that still managed to pull sales above 2M, the majority of them reportedly in the first couple of months, so at full price. 2B has flight attacks, and has a special evade mechanic that can quickly move her to an advantageous position. Many of these include a new icon on the UI. Then, I dislike that regardless of the installation dir, an INI file is written to C: root and each time I start the game, some file is copied from the CD to the HD, duh. I'd like to hear from others about this oddity. It still requires the CD-ROM but at least it works though it uses 19MB instead of 8MB disk space. I worked around it by installing the entire CD-ROM PINBALL subdir in the root of the install partition. Unless there is a unique problem with my CD-ROM reader or the drivers, it almost looks like they screwed up with relative path references inside an executable. I could start the menu but not get to any tables. To avoid too much detail, the standard installation refused to find certain data files on the CD-ROM. I had a particular installation problem with the MS-DOS version and the install instructions are contradictive in a few places. SFX are cheap and few while the fluffy synthesizer music reminds one of Leisure Suit Larry walking through hotels and riding elevators. Sound-wise, both the effects and the background tunes are marginal. An absolute faux-pas for something that wants to be called a simulation and advertises "authentic ball movement to perfection" on the box. Also, unless the ball directly hits the pivot, it can't be stopped and cradled, it rolls right off the flippers. The slingshots are somewhat set back, exposing a piece of the in-lane rails to direct hits where the ball bounces off too hard and usually arches outside the reach of the flippers - it's right there but you can't reach it and have to wait for the trajectory to complete. In the flipper area it gets downright annoying. The ball generally moves softly and slowly, unnatural for a steel ball, making gameplay somewhat unnerving and boring soon. I'm especially disappointed in the physics again. It's got crisper graphics, more complex layouts and more advanced features than the dated Fantasies but doesn't top the non-scrolling Pinball Illusions (still the best 21st game, IMHO) in either originality, excitement or ball physics. Quality-wise, Absolute falls somewhere in between the likes of Pinball Fantasies and Pinball Illusions with a hint of Psycho Pinball. A lot of effort has been put into the enjoyable dot-matrix animations. Multiball here is rather unexciting and anyway, why bother? Why wouldn't I already play in the most optimal resolution during single ball? Various video modes are featured that involve the usual games of hand-eye coordination skills. The only other available game option aside from sound volume is a seperate resolution for multiball play. However, Absolute considerately supports the Thrustmaster Pinball Wizzard (even in DOS) and the Philips Virtual Pinball Controllers (which I do not own). All keys are set and can't be customized. Screen resolutions can be switched on-the-fly via keys 1-4. Tables can be nudged left, right and up, tilt seems to be rather insensitive. Highscores can be reset but default back to annoying preset scores. All games are 5-ball play with the top 5 scores being recorded. Space is taken up by the dot-matrix display situated at the bottom end of the tables. A clever idea but how many people really feel comfortable doing this each time? Unfortunately then, the next best and preferred resolution is the scrolling "VESA Norm" which manages to display about 2/3 of a table at a time. This is the only non-scrolling display but requires the monitor to be put on its side. The last one is called "VESA Flip" and rotates the tables 90 degrees. The 2D scrolling planview can be employed in three VGA and two VESA resolutions: 320x240, 360x270, 320x400, 640x480 and 480圆40. It features four different tables whose themes - Paris-Dakar rally, Hollywood, deep sea, baseball - are not entirely original but sufficiently different from what has been done before. Absolute was designed by UDS - Unique Development Sweden - a growing company that got its start in the Atari world. Whether or not they were made with such portentous meanings in mind, it's easy to find meditations on faith, belief, devotion, life, and death hiding within the narratives of these animated classics. However, that doesn't mean such readings are unearned. Finding deep adult themes in the Toy Story films has been a favorite past time of many a critic and cinephile since the first movie was released back in 1995. The reason for my morbid remembrance is that few films have ever captured the dreaded feeling that comes with the confrontation of one's mortality as well as the third cinematic adventure of Woody and Buzz. I lived, but I'll always remember the feeling of thinking I was going to die, the fear, and the resigned acceptance of it.Īnyway, let's talk about Toy Story 3 on its 10th anniversary… I couldn't breathe, I was alone and started to lose consciousness from lack of oxygen, gasping for air while the world around me was going dark. It was late at night, I was eating something and a piece of food got stuck in my throat. The third moment where I contemplated my death in a very immediate way is, weirdly enough, the one that still scares me the most. From the most searing pain I've ever felt to internal bleeding after surgery, it all seemed like it was going to end. The second time was considerably less spectacular, caused by gallbladder stones and some incredible bouts of bad luck. The first was in 2011, in Tokyo when the Tōhoku earthquake happened, making me stare in horror as skyscrapers swiveled around me, looking like they could fall at any moment. Have you ever thought to yourself "my time has come to an end, I'm going to die"? I have, in at least three instances. 3/29/2024 0 Comments Fireside BowlIt wasn’t until a few months later that I actually opened and read my stolen find. I casually slipped the zine into my back pocket and walked away, rejoining the crowd to watch the rest of the show. Surveying the room I noted everyone else’s interest was being funneled toward the stage opposite me. I knew I had to have it, but I’d already spent the last of my money at the door. The dog-eared pages were already starting to let go of the staples binding them together, giving off a wafting history of many good reads. The yellow cover had been softened from use and held the blanket texture of long circulated dollar bills or paper having been run through the washing machine. It was the eighth issue, particularly thick, with a nice weight to it. I dug through to the bottom for something good, but the only thing to hold my attention was a tattered copy of something called Burn Collector by someone named Al Burian. There was no way any band could have competed with the experience.Īfter spending some time checking the place out I came across an abandoned box of zines at a merch table in the back. Walking into the Fireside Bowl that night was the urban equivalent of repelling down into some long lost geodesic cave. Still young to the city I was a fresh receptor to its varied stimuli, its mysteries shedding themselves to me daily and by the minute. I was, however, thrilled by the building itself. It was my first time and really I was pretty disinterested in the bands on stage, most of which I had probably never heard of at the time. I’d been living in Chicago for about a year before my friend Jared took me with him to a Fireside show. R.I.P.Īpril 6th, originally uploaded by rebecca ann rakstad. Thanks for all the wonderful memories mister Fireside Bowl. That will be shut down probably by the time you’re reading this. But that’s a whole other band and a whole different all ages venue. After all these years of going to show I can honestly say that I have only seen one better show than that. The band did an encore and I knew that I had just seen the best show in my life. I didn’t even think that was possible and too this day I haven’t experience that again. My friend yelled out to me “it’s so hot in here my eye lids are sweating.” I realized that mine were too. I don’t know how my body was surviving all the heat. The guitar player ripped down a tile from the ceiling with his pointy guitar head. The band just kept tearing through song after song. He turned to me and said, “this is the best night of my life!” wow, it felt so good to be alive. The guitar player was swinging his guitar around so much that he sliced a kids face open. They put danger back into Rock n’ Roll that night (which is missing a lot these days but to me is a main part of rock). The kids went fucking crazy! I had never seen anything like it. They hit the stage and opened with Panasonic Youth, a new song. They finished and I made my way up to the front to see who I came to see, The Dillinger Escape Plan. Fucking awesome! The rest of the set was totally amazing. He wondered “what the fuck are you doing in the dirty ass Fireside Bowl bathroom when there are hundreds of bbq’s going down outside.” He then dedicated the next song to the fly. After one of the songs the singer talked about taking a piss and seeing a fly in the bathroom. The kids went wild! Everyone was dancing and going nuts. The next band was The Bronx, who totally fucking slayed everyone’s faces off. It was sold out and everywhere I looked someone was smoking a cigarette. It had to have been 100 degrees in there. What a fucking dickhead! By then the heat was starting to get to me. A kid right behind me was waving a big piece around until a bouncer came and kicked him out. They threw the broken pieces into the audience, who started smashing them even more. At the end of their set they picked up two laptops, smashed them and talked about how technology was destroying music. Next up was Decahedron, who was so much better. Great name but what a shitty band! A pit had already started. SWEET! We got in and soon Planes Mistaken For Stars hit the stage. My friends’ mom let us borrow her credit card to get tickets online so we didn’t have to worry about it selling out. We arrived to the show and there was already a huge line. No booze or dope for the night but I didn’t even care, I was just happy to get the hell out of work. My two best friends picked me up and we headed straight for the Fireside. It was a long day of picking up dog and cat shit at the shelter. There was a continued emphasis on the running game in 2000 – although the approach became a running back-by-committee one. His game-plan against the Jets in 1999 was what I thought his blueprint in New York would be – a heavy dose of a feature back (in that game, Joe Montgomery carried the ball 38 times) and a lot of play-action. To his credit, Fassel admitted to the inadequacy and promoted Payton. Payton, you will remember, was elevated to the offensive coordinator position when it became obvious that Fassel was having difficulty calling plays and managing the game at the same time. But unfortunately, that’s not Jim’s nature.įassel’s biggest coaching problem this year has been Offensive Coordinator Sean Payton. Personally, I wish Fassel had more of a kick-butt personality about him in spirit of a Jon Gruden as I think players will reflect the nature of their head coach. Fire Fassel and you could get lucky and find another Bill Cowher. Unless a coach has a special aura about him such as a Bill Parcells, selecting any new coach is a crap shoot. Once a player signs a big contract, it becomes more and more difficult to motivate him – it’s simply human nature. With ever growing player salaries/bonuses, the salary cap, and free agency, players have more leverage vis a vis the head coach than they ever had. But before you start yelling for firing Fassel and hiring a guy who will instill fear, note that those kind of guys are difficult to find. They don’t really fear him and thus his threats to punish are most likely not taken that seriously. Fassel’s major con is that he is a good guy and his players know it. Fassel’s players like him and continue to play hard for him. Many coaches do not have the latter asset as most are stubborn to the core. Fassel’s assets are that he is very intelligent, analytical, organized, hard-working, and somewhat open-minded. Bad coaches don’t win two division titles, a conference championship, and a “Coach of the Year” award in their first four seasons. He’s not a great coach, but he’s not a bad coach. Let’s break down our analysis into the following compartments: coaching, talent evaluation, injuries, players and 2001-specific factors. It’s not just the coaching, the injuries, the schedule, or the players – but a combination. There is no easy scapegoat to pick on here. Game Overview: What is going on? How can the Giants be in the Super Bowl one year and be fighting to stay out of last place in the NFC East just one year later with virtually the same roster? Instead of reviewing the unit-by-unit breakdowns again (something that would sound like a broken record in any event), I want to use the review this week to address the various interrelated factors that have contributed to the Giants’ demise in 2001. |
